Bob is miserable due to his 25 years long marriage issues. Bob is staying at Park Hyatt Tokyo.

Charlotte is a person that studied philosophy at university. Charlotte has travelled with her husband John to Tokyo because he is working as a photographer. Charlotte has been married to John for two years. Charlotte is also staying at Park Hyatt Tokyo, like Bob. Charlotte and Bob become friends.

An actress in Hollywood, Kelly annoys Charlotte. Kelly is a ditzy actress. Kelly is staying at the same hotel, to promote a film. Kelly sees Charlotte and John. Kelly starts talking about photo shoots that were done by John. Bob and Charlotte meet each other at times at the five star hotel, eventually interacting at the bar. Soon John goes away outside Tokyo for his assignment. Charlotte invites Bob to explore around Tokyo during the night, they end up singing at a Karaoke box. After days, one night, Bob and Charlotte see television and drink sake. They discuss a personal discussion about Charlotte's doubts. They talk about Charlotte's failing marriage. They also discuss Bob's failing marriage.

Another day, Bob and Charlotte eat a shabu-shabu lunch together. Later at night Bob tells Charlotte he will be leaving Tokyo the next day. Bob and Charlotte tell each other how much they will miss each other. They go to the hotel bar for a final social event. The next morning is the ending of Lost in Translation. Bob and Charlotte say goodbye to each other. In the end Bob kisses Charlotte before he departs.

The essence of this film is that it shows the experiences of Bob and Charlotte in Japan.
